It features a LC4c single-cylinder, DOHC, 4-stroke, liquid-cooled engine with a displacement of 398.7cc, producing 44.2 hp at 8500 rpm and 28.8 lb-ft of torque at 7000 rpm. The bike has a 6-speed transmission with a slip/assist clutch and a quickshifter as an option. It also includes a 4.2-inch TFT display with Bluetooth connectivity for music, incoming calls, and turn-by-turn navigation.

The chassis consists of a two-piece steel trellis frame with a different steering head angle, subframe mounts, and rigidity profile compared to the 390 Duke, designed to improve handling, control, and stability. The front suspension includes a 43mm WP Apex fork with adjustable compression and rebound damping, while the rear suspension has a fully adjustable WP Apex monoshock. Both the front and rear suspensions offer 9.05 inches of travel.

The bike comes with two ride modes: Street and Sport, and the Motorcycle Traction Control can be turned off. The front brake is a 4-piston radial-mount caliper with a 320mm disc, and the rear brake is a single-piston floating caliper with a 240mm disc, both with ABS. The bike is equipped with Michelin Power 6 tires, 110/70-17 at the front and 150/60-17 at the rear, on wire-spoked wheels.
